INTENDED USE
Qualitative detection of Entamoeba histolytica antigen in feces collected from veterinary patients, for veterinary point-of-care use. It aids in the diagnosis of amoebic enteritis, which can cause dysentery and chronic diarrhea, particularly in dogs, cats, and nonhuman primates. Results must be interpreted together with clinical signs, fecal microscopy, and other diagnostics, noting that antigen detection reflects active intestinal infection while morphologically similar non-pathogenic species are not distinguished by microscopy alone.
PRINCIPLE
The Amoeba Ag Rapid Test is a lateral-flow immunochromatographic assay based on colloidal gold labeling. The test cassette contains a nitrocellulose membrane with a test (T) line pre-coated with capture antibody and a control (C) line. When the processed fecal sample is added to the sample well, it migrates by capillary action. If Entamoeba histolytica antigen is present, it binds the gold-conjugated antibody and is captured at the T line, forming a visible colored band. The C line should always appear, confirming the assay ran correctly. Results are read visually by the naked eye within the stated time; no instrument is required.
SPECIMEN
Use a fresh fecal sample from the animal. Collect the sample and process it promptly, as amoebic trophozoites degrade quickly once outside the body. Mix the feces well with the provided assay buffer according to the kit procedure and test without prolonged delay. Avoid desiccated or heavily contaminated samples, which may reduce reliability.
